The joint effects of arsenic exposure, cigarette smoking and TNF-alpha polymorphism on urothelial carcinoma risk

摘要

The aim of the present study is to investigate the combined effects of urinary total arsenic level, cigarette smoking and secondhand smoke on Urothelial Carcinoma (UC) risk. We conducted a hospital-based case-control study consisting of 584 UC cases and 1098 cancer-free controls and found that ever smokers who exposed to secondhand smoke and carried the G/A and A/A genotypes of the TNF-α -308 G/A polymorphism had a significantly higher UC risk (OR = 5.17, 95% CI = 2.26–11.86). Ever smokers who exposed to higher total arsenic level (≥ 16.38 μg/g creatinine) and secondhand smoke had a significantly increased UC risk (OR = 13.18, 95% CI = 6.48–26.83). In conclusion, our findings suggest that the joint effects of urinary total arsenic level, cigarette smoking, secondhand smoke and the TNF-α -308 G/A and A/A genotypes on UC risk were statistically significant and showed a dose-response relationship.
