The interactions between avian H5N1 influenza virus and dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN)

Dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN) has been identified as a receptor for human immunodeficiency virus type 1, hepatitis C virus, Ebola virus, cytomegalovirus, dengue virus, and the SARS coronavirus. In this chapter, we discuss experiments on the interactions between DC-SIGN and H5N1 viruses using pseudotyped and reverse-genetics (RG) technologies. In addition, electronic microscopy and functional assay were employed to elucidate the morphology and function of pseudotyped viruses containing hemagglutinin (HA) and neuraminidase (NA) proteins from H5N1 strains. Results from a capture assay showed that DC-SIGN-expressing cells as well as dendritic cells from human peripheral blood are capable of transferring H5N1 pseudotyped and RG virus particles to target cells; this action can be blocked by anti-DCSIGN monoclonal antibodies. In summary, DC-SIGN mediates infections both in cis and in trans for avian H5N1 virus transmission.
