LMP1 gene detection through polymerase chain reaction in microfluidic chip combined with nanoslit surface plasmon resonance sensor

摘要

Latent membrane protein 1 (LMP1), one of the main performance proteins of the Epstein-Barr virus (EBV), also highly associated with Nasopharyngeal carcinoma (NPC). Herein, a combined microfluidic polymerase chain reaction (PCR) device with nanoslit surface plasmon resonance (SPR) sensor for LMP1 DNA detection has developed, enabling the amplification and detection of the LMP1 gene on a combined platform. With 30 cycle PCR amplification, the detection limit of 10 pg/mL can be found. Moreover, extracted DNA from EBV-positive cells could be detected through the device, showing the potential of clinical diagnosis. This integrated device performed a label-free, real-time, highly sensitive, low cost, and rapid detection.
