A HIGH-THROUGHPUT-3D COLON CANCER TUMOR MICRO-SPHEROIDS FOR STUDYING TUMOUR MICROENVIRONMENT INDUCED STIFFNESS VARIATION AND DRUG RESISTANCE

摘要

Fibroblasts are the key components of tumor microenvironment (TME) and activity of these cells induces desmoplastic reactions in the spheroids. In this present work we co-cultured colon cancer cells (HCT-8) and fibroblast cells (NIH3T3) in a micro-well 3D (µ- well 3D) array cell culture chip and produced high throughput, even sized multicellular spheroids. The PDMS and contained 630 round wells, were coated with Pluroninc F-127 self-assembled molecules for rapid single organoids growth. The cross talk between colon cancer cells and fibroblast cells upregulated cancer associated fibroblast (CAF's) markers, which are co-related with increased stiffness and drug resistance in 3D co-cultured spheroids.
