Peritoneal Tumor Microenvironment Labchip For The Selection Of Hipec Drugs

Abstract

Cancer is the leading cause of death worldwide, and therapies to overcome cancer metastasis and cell proliferation are challenging. Hyperthermic Intraperitoneal Chemotherapy (HIPEC) is often a last resort. Our goal is to develop a platform for HIPEC drug screening. We have fabricated a three-layer microfluidic chip with specific channels designed for the automatic combination of drugs. Additionally, cancer cells and fibroblasts are arrayed to mimic the actual tumor microenvironment. Hydrogel and collagen were used for cell attachment, and drugs flowed directly to the cell culture chamber. During the experiment, the operating temperature was maintained at 43 . Finally, cells were stained with the LIVE/DEAD assay to observe cell viability. Based on the experimental results, the combined effect of two drugs is higher than that of single drug conditions. The combination of Docetaxel and Mitomycin C had the most significant impact among the various drug tests used in this research study.
