N2 - OBJECTIVE: Reports show that microRNAs polymorphism is related to the genetic susceptibility to lung cancer,while the conclusions are inconsistent.In this study,meta analysis was performed on related literatures published from January 2007 to April 2017 to evaluate the association between polymorphism at miRNA-196a2 rs11614913 and miRNA-146a rs2910164 and genetic susceptibility to lung cancer.METHODS: The databases of PubMed,CNKI,cqVIP,CBM or wanfang were retrieved to search for publications about case-control study,which studied on the relationship between rs11614913 or rs2910164 polymorphisms of miRNA and genetic susceptibility to lung cancer.According to the inclusion criteria and exclusion criteria,relevant data were extracted from qualified literature.RevMan5.3 software was used to perform the heterogeneity test on the results of the selected literature,the odds ratio(OR) and confidence interval(95%CI) were calculated after the heterogeneity test.The funnel plots were protracted to estimate the effect of publication bias Meanwhile,subgroup analysis was performed according to the difference in races and source of control.RESULTS: Totally 9 eligible articles were included and analyzed.There were 3 101 cases and 3 234 controls for miRNA-196a2 rs11614913 and 3 483 cases and 3 578 controls for miRNA-146a rs2910164.In the combined study,miRNA-196a2 rs11614913 was observed to be significantly associated with lung cancer risk(CC vs TT:OR=1.25,95%CI:1.09-1.44,χ2=10.03,P=0.12; CT vs TT:OR=1.26,95%CI:1.03-1.53,χ2=13.07,P=0.04).Subgroup analysis revealed that CC and CT genotypes were associated with increased susceptibility to lung cancer in Asian populations; CT genotypes in hospital controls were associated with increased susceptibility.For miRNA-146a rs2910164,only the CC genotype was found to be associated with increased lung cancer risk (OR=1.30,95%CI:1.13-1.49,χ2=2.64,P=0.75).Subgroup analysis showed that the CC genotype of the Asian population was associated with an increased susceptibility to lung cancer,and the CG genotype of the Caucasians was associated with an increased susceptibility to lung cancer.The CC genotype in both the community and hospital control subgroups was associated with increased susceptibility to lung cancer.CONCLUSION: The genotype CC or CT of the miRNA-196a2 rs11614913 and the CC of the miRNA-146a rs2910164 may increase the risk of lung cancer in the population.
