Description
Ulva prolifera is a kind of common wild green algae in Taiwan, with its rich resource and nutrient, food and medicinal value. Previous studies have extracted several polysaccharides from Ulva prolifera and shown their bioactivities, including antioxidant, anticoagulant, and immunomodulating effects. In addition, some other studies indicated that polysaccharides extracted from Ulva prolifera reveal anti-cancer properties. In our preliminary results, we found that extracts from Ulva prolifera could specifically inhibit the growth of colon cancer cell lines without affecting nonmalignant cell lines, and their inhibition was correlated to the content of polysaccharides. Hence, in this plan, we purpose to investigate the effects of polysaccharides extracted from Ulva prolifera on malignant characteristics of colon cancer precisely. Furthermore, we expect to develop polysaccharides extracted from Ulva prolifera as a novel alternative and complementary medicine for future colon cancer treatment. Specific aim 1: Optimization of polysaccharide extraction of Ulva prolifera. Specific aim 2: Investigation of the effects of polysaccharides extracted from Ulva prolifera on the malignant characteristics of colon cancer. Specific aim 3: Identification of the underlying mechanisms and signaling pathways involved in the inhibition of polysaccharides extracted from Ulva prolifera on colon cancer. Specific aim 4: In vivo evaluation of anti-cancer effect of polysaccharides extracted from Ulva prolifera. In vivo pharmacological selection for potential anti-cancer agents will be one of the most important goals in this proposal. We also purpose to translate the research consequent of this proposed project from basic research to possible clinical trials.
