The metabolic syndrome causes a considerable burden on health and social care,and the study of its causes has become an irreversible trend. Endothelial progenitor cells play an important role in the repair of vascular endothelium and vascular hyperplasia during hypoxia. Additionally, recently studies have shown that EndoMT plays crucial physiological and pathophysiological roles in regulation of endothelial function, the development and structural remodel of cardiac muscle, cardiac valve and blood vessels. This study aimed to investigate the concentration-related effects of HDL and oxLDL on in vitro and in vivo function of human endothelial progenitor cells (EPCs) and related EndoMT. May be able to clarify the mechenism on EPCs function an endoMT through clinical observation and in vitro study. We also believe the results of our study will provide the more effective treatment protocols for patients with dyslipidemia in the future. which may provide novel targets for the prevention and treatment of cardiovascular diseases.The project is expected to complete a number of objectives in two yearsFirst year: The specific aims are as follows: (Part I) 1. To observe the effect of TGF-b on EC/EPC EndoMT.2. To observe the lipoprotein from health people regulation of cell EndoMT. 3. To observe the effects of risk factor on cell EndoMT progress. 4.Create animal model for second year study. (PartII) The effects of lipids on angiogenesis and EndoMT were confirmed by animal experiment1. To anylsis the effects of lipoproteins from healthy individuals on endothelium regeneration and endothelial progenitor cell function in endothelium-denuded mice. 2. Comparison of effects of normal HDL and defective HDL on EndoMT in mice. 3.To investigate the effect of simvastatin on endothelial-mesenchymal transition induced by defective lipoprotein.Second year: clinical observation and data analysis as follows: 1. To observe the different patient's blood high-density lipoprotein antioxidant capacity and the impact of EndoMT. 2.To observe the effect of cardiac catheter stenting on EPC function and EndoMT in patients. 3.Analysis of the correlation between circulating EPCs and carotid ultrasonography in heart cath patients
