Description
The aim of this study is to evaluate the effect of light-emitting diode (LED) light on Porphyromonas gingivalis (P.g) in periodontitis for anti-inflammation and alveolar bone regeneration. Previously, we have established the ovariectomized senescence-accelerated mice (OVX-SAMP8) animal model for osteoporosis regeneration, and demonstrated strong recovery of osteoporosis by stem cell and platelet-rich plasma (PRP)(Biomaterials, 2011 and 2012). Based on this preliminary data, we will address two major issues in this proposal. Specific aim 1: we will investigate the effect of LED on anti-inflammation and alveolar bone regeneration. First, we will create the cell model of anti-inflammation in LED treated fibroblasts after P.g infection. Second, we infected osteoblasts with P.g to observe the osteogenic differentiation after LED treatment. Therefore, in order to better recapitulate the dynamic interactions between fibroblasts and osteoblasts within the periodontal microenvironment, fibroblasts and osteoblasts will be co-cultured under P.g infectious medium, and we will examine the role of LED in inhibiting inflammation and promoting osteogenesis in a fibroblasts/osteoblasts co-culture system. Specific aim 2: we will investigate the effects of LED on anti-inflammation and alveolar bone regeneration on periodontal mice, and this aim will create two major animal models. First, prevention model, mice will be induced alveolar bone loss and also treated the LED by oral infection with P.g at the same time. Second, therapeutic model, mice will be induced alveolar bone loss after oral infection, then, using the LED to treat the mice of alveolar bone loss. Thus, this proposal will combine these two strategies including anti-inflammation and bone regeneration to address fundamental issues in oral infection with P.g, and may be applied in the prevention and treatment of periodontitis.
