營養過剩會導致白色脂肪組織擴張和脂肪細胞缺氧,誘發免疫細胞浸潤和炎症,減弱脂肪細胞褐變,降低能量轉換, 惡化新陳代謝,促進全身炎症, 導致心血管代謝疾病。本計畫旨在研究C-C 基序趨化因子配體4(CCL4),一種新型趨化因子,在脂肪細胞炎症和褐變中的作用, 除以臨床研究確認CCL4和人體褐色脂肪組織功能相關, 並以脂肪細胞特定基因剔除小鼠驗證:直接抑制CCL4可否改善脂肪細胞炎症和脂肪細胞褐變, 並減輕高脂肪飼料餵食所導致之胰島素抵抗高血壓和微血管生成受損, 以做為防治肥胖相關心血管代謝疾病的新策略。此外,一種新型小分子的CCL4直接抑製劑將被用於驗證支持未來更多以CCL4為靶點的新防治策略。
