In recent years, many literatures demonstrate that cell sheet engineering is suitable culture technology for squamous epithelial tissue, so we started related studies from 3 years ago. We fabricated a series of electrospun polycarbonate membrane and graft-polymerized with NIPAAm as an insert of culture well for promotion of epithelial cell culture efficiency. The results plays that cell sheet could be successfully cultured and be detached. However, the residual NIPAAm monomer is not easy to clean lead to cell toxicity in the fabrication process, and the cultivation process is difficult to monitor due to the opacity of electrospun membrane process in response to the film difficult to remove residual NIPAAm monomer. Therefore, to develop a safe and available cell sheet engineering systems is very important for tissue engineering. The major purpose of this grant is to develop a feasible in non-pNIPAAm of Cell Sheet Engineering System. Briefly, Tissue Culture Polystyrene, Polyethylene and Polyvinyl difluoride will be used as a matrix and it will treat by plasma, crosslink with PGA by cysteine or cystamine. The obtained membrane will be evaluated to form a cell sheet by various cells. We hope the novel cell sheet engineering system could effectively provide a pre-ECM containing cell sheet for the cultivation of autologous tissue in the near future.
