乳癌是抗藥性與復發率很高的癌症,加上癌細胞中CDK16蛋白質激酶的變異及高度活化,導致病患的高死亡率。本計畫將透過蛋白質晶體學技術與計算模型策略,針對CDK16抑制劑進行活性最適化,以設計具有高活性與高選擇性之新穎性小分子藥物;將抑制劑針對乳癌細胞和動物模型進行抗癌藥效機轉與藥毒理研究,驗證抑制劑之抗癌療效。應用蛋白質晶體學技術與計算模型設計抑制劑能大幅縮短藥物至臨床前試驗的研發時程和降低開發成本,希冀未來能夠造福乳癌病患,提升病患的治療效果與生活品質。
