Evaluating the Role of Fibrinolytic Proteins on Insulin Secretion and Regeneration of Islet Cells in Obesity and Type 2 Diabetes during Exercise

In recent years, the population of patients with type 2 diabetes rapidly boost with increasing prevalence of obesity in worldwide. Obesity and type 2 diabetes mellitus related complications. Many large clinical trials proved that treatment of lifestyle intervention prevent type 2 diabetes and delayed development of diabetic complications effectively. Impaired insulin secretion and insulin resistance are two important characteristics in type 2 diabetes mellitus. Besides of improving heart and lung functions, regular exercise decreases insulin resistance effectively. Thereafter, exercise plays an important role on treating obesity and type 2 diabetes mellitus.Urokinase plasminogen activator is well known to be fibrinolytic proteins and wildly used on treating cerebral infarction and peripheral arterial occlusive disease. Except of fibrinolysis, urokinase plasminogen activator contribute to regulation of immune cells, cellular repairmen and remodeling. In addition, urokinase plasminogen activator takes part in process of atherosclerosis. In our previous researches showed deficiency of urokinase plasminogen activator impaired insulin secretion and regeneration of islet cells which was linked to development of type 2 diabetes mellitus. Some studies indicated increasing urokinase plasminogen activator level during exercise. We found exercise increased circulating irisin in preliminary results of our first-year experiment. In addition, FNDC5 expression in pancreas, muscle, brown and white adipose tissue showed different changes in uPA -/- mice. In the present study, we explored the role of urokinase plasminogen activator on insulin secretion and regeneration of islet cells during exercise.
