According to the previous studies and our preliminary data, liquid diet with high percentage saturated fatty acid decreased fat accumulation in liver but lead to hepatic fibrosis in rats under chronic ethanol feeding. Besides saturated fatty acid from animals and vegetables may play different role in the alcoholic liver disease (ALD). Therefore we are going to carry out the following three-year study to clarify how the different source of saturated fatty acid works in ALD. In the first year, the effect of saturated fatty acid on rats under chronic ethanol feeding based on lipid metabolism, oxidative stress, and the relationship with cardiovascular disease will be investigated. In the second year, we will study the effect of saturated fatty acid on rats under chronic ethanol feeding focusing on immune system. In the third year, we will investigate the effect of saturated fatty acid on rat under chronic ethanol feeding based on intestinal health, including fecal microbial and intestinal permeability. In the first year, we expect that both lard and cocoa oil increase oxidative stress and the level of cell adhesion molecules, thus increase the risk of cardiovascular disease in ALD. In the second year, we expect that both lard and cocoa oil induce inflammatory response in rats under chronic ethanol feeding and enhance proinflammation cytokines concentrations in vitro. In the third year, we expect that both lard and cocoa oil contribute to intestinal damage such as change the fecal microbial and increase intestinal permeability and cause endotoxemia.
