Effects of Onion and Allium Vegetables Cultivated in Taiwan and Mongolia on Osteoporosis

Osteoporosis characterized by the loss of bone mass and microstructure, and it is associated with an imbalance of the bone remodeling process which includes bone resorption by osteoclasts and bone mineralization by osteoblasts. It has been indicated that osteoporosis is associated with the oxidative stress and inflammatory responses, so that foods or compounds that suppress such responses may possess protective effects. Allium vegetables, including onions (Allium cepa L.) and garlic (Allium sativum), are one of the widely consumed spice vegetables in Taiwan and Mongolia, and have been shown to possess antioxidative, anti-inflammatory, and anti-osteoporosis activities. There are more than 30 species of Allium species plants grown in Mongolia. Due to the harsh climate conditions with extreme differences on the temperature, the contents of the active compounds in the plants may vary. Previously, studies have showed that onion Allium cepa L. decreased osteoporosis in ovariectomy (OVX)-induced animals, but the effects of other Allium vegetables, the active components, and the mechanisms involved have not been determined and identified. Therefore, this study is aimed at screening and evaluating the potential protective effects of onions and other Allium vegetables cultivated in Taiwan and Mongolia on osteoporosis by using both ovarietomized animals and cultured osteoblasts and osteoclasts. In the first year, we will perform the extraction, and the extracts of the vegetables will be screened in vitro and in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages for their anti-oxidative activities and anti-inflammatory activities, respectively. In the second year, the vegetables of the most active extracts will be used to examine the effects on OVX-induced osteoporosis animals. In the third year, the cultured osteoclasts and osteoblasts will be used to study the mechanisms of the anti-osteoporosis effects of active onion extracts. Results obtained from these experiments may provide information for understanding the mechanisms through which the onions cultured in Taiwan and Mongolia play a role in osteoporosis.
