Description
The purpose of this project is to develop a tissue engineering complexes for the large bone defects in clinical treatment. In this study, we would like to design a 3D pore-connected calcium phosphate (3D-PCP) block, and using microgravity bioreactor cultured method enhance human dental pulp stem cells (hDPSCs) growth in 3D-PCP, finally, platelet rich fibrin (PRF) is integrated in system for providing autologous growth factor.We hope that can replace the use of autologous bone graft. We proposed a three-year project. The major task of first year includes developing a stable and porous controllable 3D-PCP process by phenomenon of thermo-expanded PEC/Bioceramic and establishing hDPSCs separation, purification, and identification protocol. In the second year of project, various 3D-PCP are cultured with hDPSC by microgravity bioreactor, that is in order to estimate the influence of various pore sizes for cell attachment and proliferation, and then the autologus PRF is introduced in previous designed 3D-PCP/hDPSCs for providing autologus growth factor. Finally, we would like to develope a bone-like tissue engineering complex for substituting autograft. In the third year’s project, the bone-like tissue engineering complex is evaluated by of Lanyu swine model with mandible bony defect that will provide prior result for IRB applying.
