胰臟癌是死亡率很高的癌症,加上癌細胞中多種蛋白質激酶,如MAP4K4、MAPK8與MAPKAPK2的變異及高度活化,導致治療的低治癒率。本計畫將透過結構為基礎之策略與人工智慧技術針對蛋白質激酶抑制劑進行活性最適化,並研發計算平台來設計高活性且高選擇性之新穎性小分子藥物;將抑制劑針對胰臟癌細胞進行細胞毒性、機轉與動物實驗,驗證蛋白質激酶抑制劑之抗癌療效。應用建立計算平台設計抑制劑,能大幅縮短藥物至臨床前試驗的研發時程和降低開發成本,希冀未來能夠造福胰臟癌病患,提升病患的治療效果與生活品質。
