Osteoarthritis is a degenerative disease of articular cartilage. Due to the lack of blood vessel, neuron, and lymph in cartilage tissue, the ability of self-repair is limited in damaged cartilage tissue. The fundamental concept of tissue engineering includes cell isolating, cell culturing in vitro, and implanting in vivo. To obtain nearly native articular cartilage tissue and repair the damaged section are currently the main purposes of cartilage tissue engineering. For regeneration of complex structural tissues, scaffolds must possess several structural features providing primary cell attachment. The specific characters such as biological compatibility, degradability, and adaptive surface chemical and mechanical properties are important. The physiographical and pathological functions of hyaluronic acid are involved widely. It can affect the embryogenesis, inflammation, wound healing, angiogenesis and tumor formation by controlling the degree of the cell proliferation, differentiation, adherence, and movement. Resveratrol is a natural polyphenolic compounds with a variety of bioactivities associated with health promotion. The polyphenolic structure of resveratrol shows antioxidant activity and may reduce oxidant-induced apoptosis and low-density lipoprotein (LDL) oxidation. Carbon nanotubes (CNTs) are a macromolecular form of carbon and are viewed as a class of nanomaterials with high potential for biomedical applications. Recent researches about the application of CNTs in tissue engineering cell scaffold have attracted tremendous attention due to their unique physical and chemical properties. Cell culture suggests that CNTs displayed high rate of cell attachment and growth in vitro culture of most kinds of cells. Higher mechanical strength is also observed after CNTs is combined with other materials. Therefore CNTs can be used for cell scaffold material of tissue engineering, and can be applied to several fields such as tissue engineering of bone, nerve, and blood vessel. The aim of this work is to fabricate and characterize the resveratrol/hyaluronic acid modified CNTs scaffolds for cartilage tissue engineering applications. The scaffold was resveratrol and hyaluronic acid modified CNTs by 1-ethyl-3-(3-dimethyl-aminopropyl) carbodiimide (EDC) and adipic dihydrazide (ADH) cross link agent. Structure, mechanical test, and in vitro degradation tests were evaluated for resveratrol and hyaluronic acid/CNTs scaffolds. Primary cultured chondrocytes, isolated from ICR mice articular cartilages, were used to confirm the cell compatibility of scaffolds. We evaluated cell attachment ratio, cell morphology, extracellular matrix synthesis, and cell proliferation to prove the potential of this scaffold to osteoarthritis therapy.
